Background: Members of the Dlx gene family contain a homeobox that is related to that of Distal-less (Dll), a gene that is expressed in the head and limbs of the developing fruit fly. The Dlx family of homeobox transcription factors (Dlx1, Dlx2, Dlx5, and Dlx6) is necessary in order for progenitors within these regions to differentiate into GABAergic neurons [1].In early mouse brain, DLX2 is expressed in neural precursor cell and neurons in the ventricular/subventricular zone of the lateral ganglionic eminence (LGE) and medial ganglionic eminence (MGE). Dlx2 promote interneuron GABA synthesis, synaptogenesis, and dendritogenesis [2-4]. Dlx2 also control neuronal versus oligodendroglial cell fate acquisition in the developing forebrain [5]. In addition, it has a critical role in craniofacial and tooth development [6, 7]. Notably, as immunohistochemistry showed its nuclear staining, DLX2 is suitable for number calculation in the CNS.
